FOXO4-DRI is built from a modified FOXO4 sequence made of D-amino acids in reverse order, which resists breakdown. It was developed to disrupt a survival interaction found in senescent cells and has been tested in mice as a senolytic.

Commonly reported reconstitution steps

  1. Vials are usually brought to room temperature (about 15-20 minutes) first.
  2. 3 mL of bacteriostatic water is added slowly down the inside wall of the 10 mg vial.
  3. The vial is swirled gently until clear, not shaken.
  4. This gives 3.33 mg/mL. Mixed vials are typically labeled with the date and refrigerated.

Storage

Before mixing: Lyophilized vials are commonly stored frozen (-20 °C) or colder.

After mixing: Reconstituted solution is commonly kept refrigerated (2-8 °C); freezing or aliquoting has not been validated to preserve it.

Commonly used within 28 days of mixing.

How it works

In senescent cells, FOXO4 holds p53 in the nucleus and keeps it from triggering cell death. The peptide competes for that binding, releasing p53 and promoting apoptosis selectively in these cells in the laboratory.

Researched for

  • Studied in mouse models of chemotoxicity and natural aging for senescent-cell clearance.
  • Examined in recent animal and cell work on fibroblast, endothelial, and testicular senescence.

Notes

  • Only preclinical data exist; animal studies used short intermittent courses rather than daily administration.
  • The daily dose steps in the reference are not derived from human trials.
