GHRP-2 (pralmorelin) belongs to the first generation of synthetic growth hormone releasing peptides developed in the 1980s-90s. It produces a strong, short-lived GH pulse and has been evaluated as a provocative test for growth hormone deficiency. It is not approved in the US.

Commonly reported reconstitution steps

  1. Vials are usually brought to room temperature (about 15-20 minutes) first.
  2. 3 mL of bacteriostatic water is added slowly down the inside wall of the 5 mg vial.
  3. The vial is swirled gently until clear, not shaken.
  4. This gives 1.67 mg/mL. Mixed vials are typically labeled with the date and refrigerated.

Storage

Before mixing: Sealed vials are commonly stored refrigerated (2-8 °C) and protected from light, or frozen for long-term storage.

After mixing: Reconstituted solution is commonly kept refrigerated (2-8 °C), protected from light, and not frozen.

Commonly used within 28 days of mixing.

How it works

GHRP-2 binds the growth hormone secretagogue (ghrelin) receptor at the pituitary and hypothalamus and acts partly through a pathway separate from GHRH, so it can amplify the response to GHRH analogs. It also raises ACTH, cortisol and prolactin to a degree in human studies and increases appetite, as ghrelin does.

Researched for

  • Evaluated as a diagnostic stimulus for GH deficiency, including in children and adults.
  • Studied for acute GH, prolactin, ACTH and cortisol responses in healthy volunteers.
  • Studied for appetite stimulation similar to ghrelin in healthy men.

Notes

  • Published human work found that the GH response lessened with repeated dosing over several days, which is why frequent dosing is a point of debate.
  • Reported effects include increased hunger and transient rises in cortisol and prolactin.
  • Reference charts list both 5 mg and 10 mg vials with the same 3 mL reconstitution; the concentration differs.
  • Developed as a diagnostic agent in Japan; no US approval.
